The PositionYou are a driven molecular biologist with a passion for developing cutting-edge technologies. Your expertise lies in functional genomics, with a specific focus on high-throughput screening approaches, molecular cloning, and next-generation sequencing analysis. You are adept at both the wet-lab execution of complex cellular assays and the computational analysis of the resulting large-scale datasets. The selected candidate will spearhead a highly innovative project to build a first-in-class CRISPR-based platform that modulates splicing to expose druggable isoforms and new splicing events, with direct line of sight to target discovery. Your work will be pivotal in translating a novel scientific concept into a powerful engine for drug discovery at Roche.
This role is part of the Roche Postdoc Fellowship (RPF) Program, which fosters frontier science and strengthens relationships with academia. You will work in a collaborative environment, bridging internal science at a Roche Innovation Centre with an academic research group to stay at the cutting edge of transcriptome engineering.
The OpportunityEstablish, validate, and optimize a CRISPR-based platform for the precise, bidirectional modulation of pre-mRNA splicing.
Design, construct, and screen complex guide-RNA libraries in pooled and arrayed formats.
Develop and leverage your computational skills to analyze large-scale genomics data (e.g., RNA-seq, BRB-seq, DRUG-seq) to identify, validate, and mechanistically characterize top screening hits and elucidate their mechanisms of action.
Work closely with a cross-functional team of scientists in Lead Discovery and Oncology, contributing to a versatile, cross-departmental discovery platform.
Drive science forward by contributing to high-impact publications and the generation of intellectual property based on the platform and its discoveries.
PhD in RNA Biology, Molecular Biology, Genomics, or a related discipline. RPF eligibility within 4 years post-PhD.
You possess a deep understanding of RNA biology, particularly pre-mRNA splicing regulation.
You have significant hands-on experience with CRISPR-based functional genomics screens; direct experience with RNA-targeting systems is a strong asset.
You are highly proficient in essential techniques such as complex cloning, lentiviral production, mammalian cell culture, and NGS library preparation.
You possess a "hybrid" skillset, combining a strong foundation in wet-lab research with an aptitude for computational genomics. Experience analyzing large-scale sequencing data is a significant advantage, and you are willing to learn and apply machine learning methods.
You are confident working independently within matrix teams and across different research disciplines, managing priorities effectively within a dynamic drug development organization.
You are fluent in English and have excellent communication and presentation skills, with the ability to convey complex scientific ideas and data in a clear and concise way, ready to contribute to a collaborative and innovative research environment.
